Hollande's fixative (DG)
A variant of Bouin's solution. It is stable and capable of decalcifying small bone specimens. Tissue fixed with Hollande's can be stained successfully with most stains, and the cupric acetate in the solution stabilises red blood cell membranes as well as cosinophil and endocrine cell granules, resulting in less lysis than with Bouin's solution. Hollande's is commonly used as a fixative for gastrointestinal biopsy tissues.
Because the salts in the solution will produce an insoluble phosphate precipitate, it is necessary to thoroughly wash the fixative prior to placing the specimen in a phosphate buffered formalin solution. Prefilled ready-to-use histo-containers are packaged in leak-proof histo-containers.
