Trypsin powder
Extract from Porcine Pancreas.
CAS 9002-07-7
Activity >100 NF units/mg
Used for whole cell preparations. (see references in Triton X-100)
How to dissolve it:
- Use an appropriate solvent:
Trypsin is most stable in a slightly acidic solution. The recommended solvent is 1 mM HCl (hydrochloric acid) to achieve a final concentration of 1 mg/mL. Alternatively, for cell culture applications, a calcium and magnesium-free balanced salt solution (like Hank's Balanced Salt Solution or HBSS) is used, often with EDTA, and the pH is adjusted to 7.4-7.6.
- Add solvent to powder: Add the appropriate amount of chilled solvent to the lyophilized trypsin powder.
- Mix gently: Swirl or mix the solution gently. Avoid vigorous vortexing, which can denature the enzyme. Gentle stirring at a low rate for about an hour is effective.
- Check for complete dissolution: The solution should appear clear, possibly with a yellow or amber tint, once completely dissolved. A small amount of precipitate may be normal, but generally, the powder should dissolve.
Storage: For long-term storage, the solution should be aliquoted and frozen at -20°C to maintain activity and avoid freeze-thaw cycles.
