Lowicryl K11M kit (DG)
Lowicryl resins are strongly crosslinked methacrylate embedding media that can be used under a variety of embedding circumstances. When they reach the appropriate hardness, they can be easily sectioned with glass or diamond knives.
Lowicryl resins are photopolymerised by ultraviolet light with a long wavelength (360nm). Since the initiation of the polymerisation is largely independent of temperatures, blocks may be polymerised at the same temperatures used for infiltration. The resin may also be chemically polymerised at 60°C. The K11M kit can be used down to -60°C and is hydrophilic.
The hydrophilic properties of K11M provide two distinct advantages.
- During dehydration and infiltration, specimens can remain somewhat hydrated.
- Better structural preservation and improved preservation of antigenicity make K11M particularly useful for immunolabelling of sections.
As with other polar (water-miscible) resins, efforts should be taken to make certain that the block face does not become wet throughout sectioning. Sections should be collected as soon as possible after they are cut, and staining incubation should be kept as brief as possible. Optimal sectioning can be done easily with uranyl acetate and Reynold's lead citrate.
