Carstairs fibrin and platelet staining, picric acid (DG)

Picric acid stain solution (reagent grade) is used in Carstairs method for Fibrin and platelets. 2,4,6-TrinitrophenolFormalin-Saline for 48 hours or moreStaining Procedures:Hydrate slides to distilled water.Mordant in Ferric Ammonium Sulfate, 5% (EMS26381-01) for 5 minutes. Rinse in running tap water.Stain with Mayer Hematoxylin (EMS26381-02) for 5 minutes. Rinse in running tap water.Stain in the Picric Acid-Orange G Solution (EMS26381-03) for 30 minutes to 1 hour. Rinse once in distilled water.Stain in Ponceau Fuchsin Solution (EMS26381-04) for 1 to 5 minutes. Rinse in distilled water.Differentiate with Phosphotungstic Acid, 1% (EMS26381-05) until the muscle is red and background pale pink. Rinse in distilled water.Stain with Aniline Blue Solution (EMS26381-06) for 30 minutes to 2 hours. Rinse in several changes of distilled water.

Formalin-Saline for 48 hours or more

Staining Procedures:

  1. Hydrate slides to distilled water.
  2. Mordant in Ferric Ammonium Sulfate, 5% (EMS26381-01) for 5 minutes. Rinse in running tap water.
  3. Stain with Mayer Hematoxylin (EMS26381-02) for 5 minutes. Rinse in running tap water.
  4. Stain in the Picric Acid-Orange G Solution (EMS26381-03) for 30 minutes to 1 hour. Rinse once in distilled water.
  5. Stain in Ponceau Fuchsin Solution (EMS26381-04) for 1 to 5 minutes. Rinse in distilled water.
  6. Differentiate with Phosphotungstic Acid, 1% (EMS26381-05) until the muscle is red and background pale pink. Rinse in distilled water.
  7. Stain with Aniline Blue Solution (EMS26381-06) for 30 minutes to 2 hours. Rinse in several changes of distilled water.
  8. Dehydrate, clear in Xylene and coverslip, using a synthetic mounting medium.
